Activation of endogenous Ras proteins by point mutations has been implicated in human carcinogenesis. We used a Ras activation system based on the expression of a transfected GTPase-deficient form of Ha-ras p21 (Delta ras-NI) and Ras-binding domain proteins (RBDs), to study the role of Ras in tumorigenesis. Ectopic expression of the transfected Ras (Delta ras-NI), but not Delta ras-NI plus a RBD, inhibited tumorigenesis in both rat and mouse fibroblast/NIH 3T3 cell systems. The ability to inhibit tumorigenesis was the result of the constitutive active nature of the endogenous Ras proteins since inhibition was observed even when expression of the transfected Ras was coincident with expression of the endogenous Ras proteins. Finally, inhibition could occur in the absence of growth factor stimulation. These results show that constitutive activation of Ras can be inhibitory to tumorigenesis and suggest that endogenous Ras proteins may function as tumor suppressors.Q:
